Decant the supernatant and resuspend the pellet in 500 μl of a suitable buffer. Transfer the solution to a Bacteria Lysis Bead Tube. Homogenize for 5 minutes at 1500 rpm (press preset button). Centrifuge for 5 minutes at 12,000 × g. Transfer the supernatant to a clean tube and store/process.   May be used for: bacteria/spores   Helpful Hints: Increase the homogenization time if obtaining low DNA concentrations from gram positive bacteria. Assays can be performed to assess optimal homogenization conditions.
